Highlightof the service
This low cost, fast turnaround, 250 ml-scale (equals to maxi prep) purification service is tailored for researcher who needs small amount of DNA for their resaerch. The service uses Qiagen’s Endo-Free plasmid purification method for plasmid less than 20 kb. Plasmid DNA purified by Express Plus quality service typically contains 0.1 EU/ug of DNA and is recommended for endotoxin-critical applications such as transfection of mammalian cells and animal-related experiments. Restriction analysis is not included in this service. Either plasmid DNA or recombinant bacteria can be submitted for this service.
Service Description
1) The recombinant bacteria we receive from the contracting laboratory
will be streaked on an agar plate with appropriate antibiotic.
After over night growth, a freshly picked colony will be used
as the starting material.
2) Unless specified by the contracting laboratory, the plasmid
DNA we receive will be transformed to DH5alpha. After over night
growth, a freshly picked colony will be used as the starting
material.
3) We will grow the recombinant bacteria using our own optimized
fermentation procedures, purify the plasmid DNA from bacteria
by column method using Qiagen's Endo-free plasmid DNA purification
kit, and dissolve the purified plasmid DNA in sterile, endofree
TE or water (<10 EU/ml). Though we do
not test the endotoxin level in every DNA preparation, our routine
quality control
tests suggest the endotoxin level in our transfection grade preparation
is about 0.01 EU/ug of plasmid DNA or less.
4) The quality of the purified plasmid DNA will be assessed by
an optical density (O.D.) ratio between 260 and 280 nM. Rrestriction
analysis is not provided in this service.
5) Five vials of glycerol stocks of the recombination bacteria,
a certificate of analysis with restriction analysis, and the
purified plasmid DNA are provided with each purification service.
Materials required for the service
Service guarantee
1) Yield:
We provide the contracting laboratory all the materials we obtain
from each preparation. Since the yield of plasmid DNA varies
depending on the nature of the plasmid, the host cell and other
factors,
we don’t guarantee the yield per liter production. We do
guarantee a minimal DNA yield of at least 100 ug/250 ml
production for low copy plasmid and 300 ug/250 ml liter production for
high copy
plasmid. In the event that the production yield is lower than
our guarantee, we will redo another production at no charge to
the contracting laboratory.
2) Quality:
O.D. ratio between 260 and 280 is at least 1.7.
Turnaroubd time for the service
3-4 days for simple project with bacteria as starting material.
4-5 days for simple project with DNA as starting material.
Additional time is required for large projects.
